Gram-negative bacilli, which are aerobic or facultative anaerobic, exhibit optimal growth at temperatures between 35°C and 37°C, with an ideal pH range of 6.8 to 7.8. These bacteria typically do not ferment lactose, except for the Arizona species. They can metabolize glucose to produce acid, although some strains like *Salmonella typhimurium* may not produce acid. Most of them generate hydrogen sulfide (H2S), do not produce a ruthenium matrix, do not liquefy gelatin, and do not break down urea. Additionally, they rarely produce acetylmethylmethanol. However, many can utilize citrate and reduce nitrate to nitrite. These organisms generally do not grow on certain selective media. The Triple Sugar Iron (TSI) test is commonly used to differentiate these bacteria. The sample is inoculated into TSI agar and incubated at 36±1°C for 18 to 24 hours. After incubation, the results are observed. In the case of *Salmonella*, the slant turns alkaline, the bottom layer becomes acidic, and H2S is produced. The reaction mechanism behind this is that *Salmonella* does not ferment lactose, so the surface of the medium remains red. However, it ferments glucose, causing the bottom of the tube to turn yellow. Furthermore, *Salmonella* can break down sulfur-containing amino acids in the medium, leading to the production of H2S. This gas reacts with lead or iron ions present in the agar, forming black precipitates such as lead sulfide or iron sulfide, which are visible as dark spots in the medium.
