**Duck Bcl-2 ELISA Kit – For the quantitative in vitro determination of Duck B-cell leukemia/lymphoma 2 concentrations in serum, plasma, cerebrospinal fluid, tissue homogenate, and other body fluids. This ELISA kit is designed to measure the concentration of Bcl-2 protein in various biological samples using a colorimetric enzyme-linked immunosorbent assay (ELISA) method. The principle of the test involves binding the target antigen (Bcl-2) to a microtiter plate coated with specific antibodies. After incubation and washing steps, a horseradish peroxidase (HRP)-conjugated secondary antibody is added, followed by a chromogenic substrate. The reaction stops with a stop solution, and the resulting color change is measured at 450 nm using a spectrophotometer. A standard curve is generated using known concentrations of Bcl-2, allowing the quantification of unknown samples based on their optical density (OD). **Sample Collection and Storage:** - **Serum:** Use a serum separator tube. Allow samples to clot for 2 hours at room temperature or overnight at 4°C before centrifuging at 2000×g for 20 minutes. Remove serum and assay immediately or store at -20°C. Avoid repeated freeze-thaw cycles. - **Plasma:** Collect using heparin as an anticoagulant. Centrifuge within 30 minutes at 2000×g, 2–8°C. Store at -20°C. - **Tissue Homogenates and Other Fluids:** Centrifuge to remove particulates. Assay immediately or store at -20°C. Ensure no hemolysis or debris is present. **Materials Required (Not Supplied):** 1. Incubator at 37°C 2. Microplate reader capable of measuring absorbance at 450 nm 3. Precision pipettes, disposable tips, and absorbent paper 4. Distilled or deionized water **Reagents Provided (Stored at 2–8°C):** - Microtiter Strip Plate (12×8 or 12×4 strips) - Standard (6 vials, 0.5 ml/vial) - Sample Diluent (6.0 ml or 3.0 ml) - HRP-Conjugate Reagent (10.0 ml or 5.0 ml) - 20X Wash Solution (25 ml or 15 ml) - Chromogen Solution A (6.0 ml or 3.0 ml) - Chromogen Solution B (6.0 ml or 3.0 ml) - Stop Solution (6.0 ml or 3.0 ml) - Closure Plate Membrane (2 pieces) - User Manual (1 copy) - Sealed Bags (1 each) **Standard Concentrations:** 200, 100, 50, 25, 12.5, 6.25 ng/mL. If sample values exceed the highest standard, dilute with sample diluent. **Important Notes:** - Do not use reagents beyond expiration date. - Do not thaw samples or reagents in water baths. - Only use reagents provided by the manufacturer. - Use deionized or distilled water only. - Keep all reagents sealed until use. - Discard disposable pipette tips after each use. - Wear gloves and lab coat during handling. - Treat all samples as potentially infectious. - Dispose of waste properly after inactivation (minimum 30 minutes). - Substrate B contains 20% acetone; keep away from heat or flame. - Allow all reagents to reach room temperature before use. **Reagent Preparation:** - Wash Solution (1X): Dilute 1 volume of 20X Wash Solution with 19 volumes of deionized or distilled water. Store at 2–8°C for up to one month. **Assay Procedure:** 1. Prepare all reagents and standards. 2. Add 50 µL of standard or sample to appropriate wells (duplicate). 3. Add 100 µL of HRP-conjugate reagent to all wells except blank. Cover and incubate at 37°C for 60 minutes. 4. Wash 4 times manually or automatically. 5. Add 50 µL of Chromogen A and 50 µL of Chromogen B to each well. Incubate for 15 minutes at 37°C, protected from light. 6. Add 50 µL Stop Solution. Color changes from blue to yellow. 7. Measure OD at 450 nm. Generate standard curve and calculate sample concentrations. **Interpretation:** - Plot average OD values against standard concentrations. - Subtract blank OD from all readings. - Locate sample OD on Y-axis and find corresponding X-axis value on the curve. - Intra-assay and inter-assay CV% < 15%. - Detection range: 6.25–200 ng/mL. Sensitivity: <1.0 ng/mL. No cross-reactivity observed. - Storage: 2–8°C (frequent use), or -20°C for long-term storage (up to 6 months). **Safety and Disposal:** - Handle all samples with care. - Follow proper biohazard protocols. - Inactivate waste for at least 30 minutes before disposal. This kit provides a reliable and accurate method for quantifying Duck Bcl-2 levels in various biological matrices, supporting research in immunology, oncology, and related fields.
