Inject the water sample or the pretreated water sample into the culture bottle. Place a carbon dioxide absorbent, such as sodium hydroxide (NaOH), into the absorption cup at the mouth of the bottle. Then, tightly seal the culture bottle and place it in a biochemical incubator. After incubating the sample at a constant temperature of 20 ± 1°C for five days, the water sample undergoes a biooxidation process under controlled stirring conditions. During this time, organic matter is converted into oxides of nitrogen, carbon, and sulfur. The carbon dioxide produced is absorbed by the sodium hydroxide, while oxygen from the headspace of the bottle is continuously supplied to meet the oxygen demand of the microorganisms in the sample. This process creates a pressure difference within the culture bottle. By measuring the height of the mercury column in the connected glass tube, the biochemical oxygen demand (BOD) of the water sample over the specified period can be accurately calculated. This method provides a reliable way to assess the organic load in water samples based on microbial activity.
